Profile
Pauline Herviou, earned her engineering degree (specializing in biotechnologies) in 2018 from AgroParisTech (Paris Institute of Life Sciences, Food and Environmental Technology). She then completed her PhD under the supervision of Dr Anne Cammas and Dr Stefania Millevoi at the Cancer Research Center of Toulouse (CRCT).
Her PhD project focused on the function of RNA G-quadruplexes (RG4) in mRNA translation and the consequences of these regulations on tumor progression and resistance to anti-cancer therapies. Her work first identified the RNA-binding proteins (RBPs) hnRNP H/F as key components of the machinery regulating RG4 structuring. She then demonstrated that these factors collaborate with the RNA helicase DHX36 to control the translation of mRNAs containing RG4s, which encode proteins involved in the cellular response to genotoxic stress. This study not only established a link between RG4-dependent translation and genetic instability but also associated this link with a role in resistance to genotoxic treatments in glioblastomas, a type of brain tumor with a very poor prognosis. Her work also identified RG4s as regulators of autophagy, a crucial mechanism for maintaining cellular homeostasis under stress and linked to tumor development and resistance to anti-cancer therapies. Overall, the results of her thesis demonstrate that RG4-RBP interactions play a central role in translational control associated with the stress response, thereby contributing to tumor development and treatment resistance.
After her PhD, she joined Prof. Martin Bushell’s laboratory at the Cancer Research UK Beatson Institute in Glasgow (UK) as a post-doctoral researcher. She is currently studying the role of eIF4A2 in controlling the localization and translation of mRNAs translated at the endoplasmic reticulum.
