Thomas Eychenne began his academic journey with a Bachelor’s degree in Chemistry-Biology from Université Paris-Est Créteil, followed by a Master’s degree in Cellular Systems Biology from ENS Paris and Université Pierre et Marie Curie.
During this Master’s program, he joined the laboratory of Michel Werner and Julie Soutourina at the CEA in Saclay to complete his internship and subsequently his PhD. In the lab, he studies a protein complex called Mediator. This complex, composed of 25 subunits in yeast and conserved across all eukaryotes, plays a central role in integrating and transmitting regulatory signals to the transcriptional machinery. The large number of subunits, some of which are essential for viability, has made it challenging to precisely understand the molecular mechanisms of Mediator action.
His work focused on studying two essential subunits of the Mediator middle module in yeast. To achieve this, he characterized temperature-sensitive mutants of the Med10 and Med7 subunits using integrative methods ranging from biochemistry to bioinformatics, including genomic approaches. He demonstrated the essential role of the middle module in establishing pre-initiation transcription complexes and its impact on the transcriptional activity of the entire genome. He highlighted a network of functional interactions between the middle module and the general transcription factor TFIIB. Additionally, He showed that the influence of Mediator varies depending on the architecture of the promoters.
He is now a postdoctoral researcher at the Institut Pasteur, where he studies the mechanisms regulating the three-dimensional architecture of chromatin and its role in transcriptional regulation.
